  • Ticket #9 – Description

    initial v1
    66....
77 5. Virus titration for infectivity
8 Each working seed lot should be assayed for infectivity in a sensitive
    9 assay in cell cultures.
10 • A plaque assay may be used in Vero or other sensitive cells. Titre
11 should be determined by counting the number of visible plaques
12 developed, and results recorded as PFU/ml.
13 • An immunofocus assay may also be used to measure virus titre. The
14 assay is based on the visualization of infected areas of a cell monolayer
15 by probing with dengue serotype-specific monoclonal antibodies.
16 Results should be recorded as FFU/ml.
17 • A tissue culture infectious dose assay may also be used to determine
18 virus titre. Results should be recorded as CCID50/ml.
